For a milligram pen: amount per click (µg) = pen total (mg) × 1000 ÷ 300. A 40 mg pen therefore delivers about 133 µg per click (40 × 1000 ÷ 300). Somatropin is filled in IU, so it reports IU ÷ 300 per click instead.

What This Reference Can and Cannot Establish

A pen click is a mechanical increment defined by a specific device. The click count, reservoir volume and labelled content are separate attributes, and none can be inferred safely from another. The geometry of one device does not transfer to another, even one that appears identical.

This reference helps a researcher check whether the required records are present and internally consistent. Delivered mass, analytical purity, identity, sterility, biological activity and suitability for a particular experiment lie outside its scope; those questions require the appropriate batch documents, analytical methods and laboratory procedures.

Verify the Physical Pen Against Its Documents

Before recording a click-derived value in a laboratory worksheet:

  1. Record the canonical product name exactly as printed on the pen and carton.
  2. Record the lot or batch identifier from the physical pen.
  3. Match that identifier to the batch-specific CoA.
  4. Confirm that the device specification explicitly states the total click count and reservoir volume.
  5. Confirm that the label and CoA use the same quantity and unit.
  6. Record the source document, revision or access date and the operator who completed the check.

Unit and Rounding Controls

Keep the source unit unchanged until the final documented conversion step. Milligrams, micrograms and international units (IU) are not interchangeable. A value expressed in IU requires a product-specific basis and must not be converted to mass without an authoritative conversion for that exact substance and reference standard. IU definitions vary by substance and reference preparation, so a generic factor would introduce unquantifiable error.

Use SI (International System of Units) symbols consistently and retain enough decimal places to show the unrounded result. The UK National Physical Laboratory’s SI units guidance is an appropriate reference for unit symbols. Apply the laboratory’s approved rounding rule only after the calculation has been independently checked.
